It's super easy and can be done with the most basic lab skills. OTOH it's probably a lot easier to just buy the yeast (unless it is a strain you have a hard time finding). I just recently started delving into legit lab work and I can honestly say that I wish I would have gotten into it years ago. It's not as hard as it seems and it is truly fascinating!

The book "Yeast: The Practical Guide to Beer Fermentation" by by Chris White and Jamil Zainasheff is excellent and will teach you how to bank and store yeast, count cells, and lots more. It's our lab manual at YHB.
